Crews is not a hardcore treatise of the subject. It does not cover the quantum mechanics behind spectroscopy and the complicated mathematical underneath the concept. Instead it discusses the fundamentals and results of such quantum calculations which apply to identification of organic compounds. Basic concepts of nuclear magnetic resonance, infrared spectroscopy, ultra violet spectroscopy are thoroughly covered with examples and charts. Crews is a splendidly-written, straight-forward work that would be suitable for advanced undergraduates who had finished at least one year of organic chemistry, for those who pursue chemical research, and entering grad students. ... This book is written by an enzymologist who presented a course on enzymology at a pharmaceutical company, as well as at the University of Pennsylvania. It is a rather well- written book that covers both theoretical and practical aspects of enzyme studies. Black and white drawings and numerous graphs illustrate the concepts. The book begins with a general review of some chemical concepts, including a brief review of thermodynamics, transition states in chemical reactions, acid-base concepts, non-covalent interactions and rates of chemical reactions. The next chapter covers aspects of enzyme (protein) structure. In a departure from other texts, the 4th chapter introduces and develops the concepts of protein-ligand binding equilibria. The derivation of Kd, along with the Langmuir isotherm to derive measurements at equilibrium is developed, as is treatment of equilibrium ligand binding data. The detour into non-enzymatic ligand-protein interactions is a very welcome treatment of this topic, so important in many drug interactions with receptors. While not strictly speaking enzymology, this chapter serves to introduce concepts that are further developed in the next chapter, which deals with the kinetics of enzymatic catalysis. This chapter nicely develops the basic equations and treatment of enzyme kinetics, and the steady state model is developed using the treatment of Briggs and Haldane. A discussion of the significance of both Km and Kcat is followed by methods to derive these values experimentally. After covering kinetics, the book moves on to chemical mechanisms of enzyme catalysis. The importance of transition state stabilization is discussed, and covalent catalysis and acid-base catalysis are highlighted. Serine proteases as model enzyme mechanisms round out the chapter.

Chapters 7 and 8 cover practical aspects of enzymology, such as velocity measurements, continuous versus end point reactions, detection methods for assays, and separation methods for reaction products (e.g., HPLC, TLC). Some pointers on enzyme concentration, temperature, pH and buffer effects on enzyme assays are valuable. The next chapter deals with reversible inhibitors, and describes the derivation of Ki. The distinctions among competitive, non-competitive and uncompetitive inhibition are discussed, and details on how to determine these in a practical sense are illustrated. There is also some discussion around the SAR of inhibitors, and this is tied in with inhibitor and drug design. Tight binding inhibitors get their own brief chapter, and there is a separate chapter on time dependent inhibition and the methodology around measuring this phenomenon. Enzyme reactions with multiple substrates and cooperativity in enzymatic catalysis also receive individual chapter treatments. Two appendices on suppliers and software tools round out the text.

I never thought this could happen to me. I picked up the book and I was hooked instantly. Never before in my forays into biochemistry have I been this enchanted by such delightful prose. I started in a thermodynamic sink and I ended as an ultra-high energy intermediate. Copeland added a section on equilibrium binding that bound me with delight. His inhibitor sections loosened all my inhibitions. Even a chowder slurping ingrate from Harvard could understand and appreciate this work.

    I have taught Biochemistry and Enzymology at the undergraduate and graduate level for over 20 years, and have recommended this book every year, to every student. Although the field of biochemistry has changed greatly during this time period, certain fundamental concepts have not changed at all, and remain central to a true understanding of how to do biochemistry. These include acid/base chemistry, energetics, enzyme kinetics, spectrophotometry, and isotopes. Of these, the first three gnerally give beginning (and even some advanced)students the most difficulty. In "Biochemical Calculations" Dr. Segel provides excellent explanations of the most important aspects of these topics. He provides example problems, with the answers worked out in detail, and then at the end of each chapter he provides numerous practice problems (along with the answer). I know of no other source of such a variety of practice problems covering these topics. Any student who expends the effort to work through these problems will certainly gain the confidence to tackle the common quantitative problems associated with biochemistry. Clearly this is not intended to be a biochemistry textbook, but rather a supplemental resource, to be used to more fully understand the topics covered in the early chapters of all current biochemistry texts.

    This is an essential book for anyone looking to practice biochemical problems -- if only for the reason that books with as many practice problems are hard to find. If you're a true biochemist and love the subject ad will continue on with graduate work or careers in biochemistry, this may be the book for you. For everyone else... probably not. This book is very minimalist. No frills. It has an answer key to the practice problems but lacks any explainations to the answer. Reviews of the material covered in the book are scant at best and very technical. Few examples from real biochemical situations in living organisms are used. It does nothing to make you interested in the subject. The book also uses units such as normality (N) and Calories (C) rather than molarity (M) and Joules (J), the more modern and internationally accepted units of biochemical equations. I have also encountered typos and mistakes in the book in my studies. The only reason why this book is still published is that there are few other books available that offer so many practice problems. If you're looking for practice on any possible biochemical calculation, buy this book. When you're done with it, compost it. Or get $2 back by selling it to the local college bookstore. Biochemical Calculations wouldn't be so bad if a new edition would be written. Biology is a rapidly changing science, and a book that is now going on 25 years old can't compete with the newer and more reader friendly texts.

    4-0 out of 5 stars A Cleverly Designed Book
    Some biochemistry books proceed to describe metabolic pathways with scant reference to the physiological context in which they occur and then fill the conceptual gap by including a chapter on regulation and integration of metabolism. Not this thoughtful and cleverly designed book. From the start the authors make it clear that they are going to tell us the story in terms of one crucial unifying concept: which pathways are activated or inactivated when we eat or when we fast?

    With this approach the reader never loses sight of the big picture, ie, what's going on in the body at any particular time. And then, when we do reach the chapter on integration of metabolism, most of the work has already been done for us: the chapter then operates as a useful review of what we have already been told.

    The book does not miss any opportunity to make us think in terms of the big picture, and furthermore, it is never afraid of saying something more than once if it needs to. Just one example to illustrate this point: Lipoprotein lipase. Action and properties of this enzyme are detailed in the context of digestion of chylomicrons (p 497), storage of TAGs in adipocytes (p 512), integration of metabolism (p 561) and the influence of insulin levels on gene expression of the enzyme (p 564).

    The early introduction of the fed and starved states is also useful in other ways. The student perceives right away what the point is of learning those endless metabolic pathways and studies them with much more enthusiasm and not just thinking that biochemistry is but an unnecessary obstacle they have to overcome to get to the more clinical stuff (at least I'd like to think so!). Inspired by this book I have organized my course in that way and I am convinced that it is the best approach.

    This book is different from its contemporaries in many ways. First of all is written in a clinical approach format which is very interesting for medical students. Most of the biochemistry textbooks are dry and vast and difficult to grasp the information needed for a medico.
    But the Marks helps to learn to use biochemistry in the process of Clinical Problem Solving. With a lot of diagrams and clear text it is easy understand.
    The Clinical Material is presented in the form of real patients.
    It is a student friendly text with which you can learn biochemistry in a funny way of solving the clinical cases.
    It can be used as a main textbook for self study.
    I strongly recommend the Marks for all medical students.

    Texts are not easily to follow. Diagrams and help notes scatters around pages which make readers confused. If you are preparing for Step 1, I recommend you study Lippincott Biochemistry, Board Review Series.

    I love this book on biochemistry simply because it was the easiest to read, with diagrams and pictures even dumb people like me can read (gg). I also found that it is very comprehensive also. Some of the other titles I've read on the subject (Lippincott, Harper's) did not go into the detail of this one. Another positive note is that this is a clinical book also; giving you case studies in each chapter to test your knowledge. I can't say enough about this book and even referred it to one of my friends. Although not required in Chiropractic College (Lippincott was), it should have been the standard. ... Read more

    This very readable book provides an excellent overview of proteomics. It gives a survey of techniques, comparing them and explaining why and when they are used. This is not the place to learn any method in detail. But it is the only book I have seen that you can read cover to cover in a few days and get the big picture of the field. Some caveats: each chapter has only 3 or 4 references, and the book focuses on methods connected to mass spectrometry. But if you want a quick and pleasant introduction to methods for purifying and identifying proteins, including discussions of ICAT, protein complexes and post-translation modifications, I highly recommend this book.

    There really are lots of good facts in this book. Somehow, though, they never come together to form a coherent whole.

    Most chapters read like essays, meant to stand on their own. That means that many chapters repeat introductory material already covered, and remake points already made. Yes, I appreciate that silver staining does not properly represent the whole range of protein expression levels. I do not appreciate the need to stat that three or four times.

    Despite its "Introduction" title, the author seems to forget what a novice will need to learn. For example, there's a discussion of isoelectric focussing for protein separation. That includes mention of the technique's history, and why it used to have reproducibility problems. I never did see an exact description of what the techique actually is or how it works, though. Likewise, the author describes a number of important software packages used in proteomics. There are no screen shots, though, comparisons of capabilities, details of usage, or even references for letting me find out more on my own.

    When I got to the the end of this book, I wanted to go back and review a few points. Because of the generally poor organization, it was always a struggle to figure out just which discussion covered the topics I wanted.
