The text should be considered for any introductory graduate level course in biochemistry. Beginning with sound chemical principles, the text lays a solid foundation for the concepts of secondary and tertiary structure within protein. The author builds a superstructure from which to view the motifs of cofactor binding domains and active sites in enzymes. Each chapter concludes with exercises, problems and "weblems". The weblems underscore the fact that structural genomics, a branch of bioinformatics, is a hot topic in the biotech arena. The weblems ask the reader to pursue ideas on the world wide web. The author provides the reader with a wealth of websites ranging from browser plug-in software for viewing crystal structures, to sources of those structures, to sequence alignment servers which will allow the student to do real research. Well thought-out, the weblems posed are useful to the student in exploring the topics of each chapter. The author sticks to protein architecture avidly, issues of how proteins fold or how structure might be predicted from amino acid sequence are presented to the reader. It is a credit to the author that he does not speculate on these very hot research topics. The bibiliography for each chapter is current to mid-2000. A student using this text will have greater insight and understanding of the literature of protein structure, folding, and prediction of structure. With this approach the reader never loses sight of the big picture, ie, what's going on in the body at any particular time. And then, when we do reach the chapter on integration of metabolism, most of the work has already been done for us: the chapter then operates as a useful review of what we have already been told. The book does not miss any opportunity to make us think in terms of the big picture, and furthermore, it is never afraid of saying something more than once if it needs to. Just one example to illustrate this point: Lipoprotein lipase. Action and properties of this enzyme are detailed in the context of digestion of chylomicrons (p 497), storage of TAGs in adipocytes (p 512), integration of metabolism (p 561) and the influence of insulin levels on gene expression of the enzyme (p 564). The early introduction of the fed and starved states is also useful in other ways. The student perceives right away what the point is of learning those endless metabolic pathways and studies them with much more enthusiasm and not just thinking that biochemistry is but an unnecessary obstacle they have to overcome to get to the more clinical stuff (at least I'd like to think so!). Inspired by this book I have organized my course in that way and I am convinced that it is the best approach.

So, you walk into a patch-clamp lab, there's a million different things on the rig and you're feeling very confused. The microscope is much more complex than the microscope you remember from high school, there's cabling literally everywhere, and everybody keeps mentioning how important it is not to disturb the "voodoo" shielding. Where do you start? What do all those instruments do? Will I ever patch a cell on my own? This is the book for you. The book does an excellent job explaining the main components on a patch-clamp rig (and advice for setting up your own if you're just starting up a lab) and gives detailed instructions on whole-cell patch clamp techniques. (I have not evaluated the single-channel techniques but they too seem quite well written.) Also, many simple "equivalent circuit" diagrams are given so that you can understand what you're measuring. You will patch successfully after reading this book! My only criticism is that the book is a bit skimpy on what to do after you've got the cell patched. Only a few voltage-clamp techniques are explored and there is very little on experiments using dual recordings. Otherwise, it's an excellent book. 1) Book is in color 2) Although best to study the book chronologically, its not impossible to skip chapters. There is a great Glossary index at the back of the page which has some definitions written into them. Furtheremore definitions seems to be clearly/concisely written throughout the book, very helpful. 3) Examples written throughout the book with soultions. 4) Parargraphs are kept to a minimum so that you wont get too bored reading through the text. I am giving this book a 4, but from my limited experience of looking at chemistry books this is the best one thats out there. It won't be a breeze studing this book, but there are many more books out there which makes the task of studying chemistry a lot harder then it is.
The book is detailed and precise. It is relatively hard yet light on math and what there is is explained well. It has great graphics and clear explanations that make the most advanced topics easy to visualize and understand. The problems at the end of the chapter are great for review. It's the best introductory chem book I've ever seen. Better than any chem course.
