An integrated and insightful look at successful drug synthesis in today’s drug discovery market

The pharmaceutical industry is unquestionably vibrant today, with drug synthesis making a vital contribution. Whether in the early developmental stages of identifying and optimizing a lead, or the latter stages of process development and cost-effective scale-up, the ability to design elegant and economical synthetic routes is often a majorfactor in the eventual viability and commercial success of a drug.

Contemporary Drug Synthesis examines how leading researchers and manufacturers have integrated chemistry, biology, pharmacokinetics, and a host of other disciplines in the creation and development of leading drugs. Authored by four of the pharmaceutical industry’s most respected scientists, this timely volume:

  • Focuses on the processes that resulted in high-profile drugs including Lipitor, Celebrex, Viagra, Gleevec, Nexium, Claritin, and over a dozen others
  • Provides an in-depth introduction to each drug, followed by a detailed account of its synthesis
  • Organizes the drugs into fourteen therapeutic areas for clarity and ease of use

Process chemists provide an essential bridge between chemistry and the marketplace, creating scientifically practical drug processes while never losing sight of the commercial viability of those processes. Contemporary Drug Synthesis meets the needs of a growing community of researchers in pharmaceutical research and development, and is both a useful guide for practicing pharmaceutical scientists and an excellent text for medicinal and organic chemistry students. ... Book Description

This second edition of a unique text/reference identifies the appearance attributes of objects and the methods available for measuring them, bringing together much material not previously organized for ready reference. The primary premise here is that ``object appearance'' involves not only color, but such attributes as gloss, luster, and translucency. The first part of the book, concerned with nature of appearance, draws from the fields of physiology and psychology and considers the eye-brain combination and the way it receives and interprets light signals. This is followed by a consideration of the optical properties of objects from the physical standpoint. The second part of the book deals with the numerical scales used to measure object appearance. The discussion here draws on psychophysics in describing the uses of physical techniques to give numbers having psychological significance. The third part of the book covers instruments for the measurement of the attributes of object appearance, their principles of design, and a survey of the major ones in use. The final chapter discusses specific applications of appearance measurement. Includes appendixes and a glossary. ... Read more

Several fundamentally important questions form the basis for this book. What are the relationships between tumour formation and tumour pH? What are the effects of tumour pH and hypoxia on carcinogenesis or tumorigenesis? What are the therapeutic consequences of tumour pH?

It is hypothesised that low extracellular pH is not only an important consequence of tumour growth but may also promote further tumorigenic transformation. Furthermore, in vitro studies suggest that low pH strongly affects the efficacy of chemo- and radiotherapy.  Better understanding of the influence of pH on tumour growth, coupled with manipulation of the pH of the tumour microenvironment, may lead to the development of more effective therapies.  ... Book Description

This book will provide a comprehensive, step-by-step approach to organic process research and development in the pharmaceutical, fine
chemical, and agricultural chemical industries. Process R&D describes the steps taken, following synthesis and evaluation, to bring key
compounds to market in a cost-effective manner. More people are being hired for work in this area as increasing numbers of drug candidates are
identified through combinatorial chemistry and high-throughput screening. The book is directed to industrial (primarily organic) chemists, and
academicians (particularly those involved in a growing number of start-up companies) and students who need insight into industrial process R&D. Current books do not describe hands-on, step-by-step, approaches to solving process development problems, including route, reagent,
and solvent selection; optimising catalytic reactions; chiral syntheses; and "green chemistry." "Practical Process Research and Development" will be a valuable resource for researchers, managers, and graduate students.

* Provides insights into generating rugged, practical, cost-effective processes for the chemical preparation of "small molecules"
* Breaks down process optimization into route, reagent and solvent selection, development of reaction conditions, workup, crystallizations and more
* Includes over 100 tips for rapid process development
* Presents guidelines for implementing and troubleshooting processes

4-0 out of 5 stars A fundamental introductory guidebook
The book starts by giving the fundamental rundown on all the major points in process development and then moves on to about twenty worked examples taken from industry. It also discusses other essential points such as economy, GMP, and the things that you don't imagine process development to be about, but should. I am relatively new to process development and I find the book very helpful, but I don't know if the book would be very revealing to a more experienced process chemist. ... Read more

Book Description

Power ultrasound has been used for many years in two specific industrial areas: cleaning and plastic welding. Over the last ten years an increasing interest has been shown in its potential for use over a much wider range of chemistry and processing which has been grouped together under the general title of sonochemistry. Most of these uses depend on the generation of acoustic cavitation in liquid media but this text, while underlining the importance of the physics and mathematics of cavitation, mainly concentrates on applications of the technology.

After an introduction to the topic and some historical background to the uses of power ultrasound the general principles of acoustic cavitation are explored including some background physics, bubble dynamics and factors which influence cavitation. The remainder of the book incorporates a series of applications of sonochemistry which illustrate the types of physical and chemical effects of ultrasonically induced cavitation which will interest chemists and engineers alike. Amongst the major topics included are chemical synthesis, environmental protection and remediation of water, sewage and soils, polymer synthesis and processing, electrochemistry including both analytical and synthetic aspects and plating. The final chapter reviews the range of ultrasonic equipment available in the laboratory and the progress made towards the scale-up of sonochemistry.

The level is introductory to semi-advanced and no topic has been taken to a particularly specialist level since it is intended that this should be of general interest to readers with a scientific background.

Book Description

The combustion phenomena of propellants and explosives are dependent on physicochemical parameters such as ingredients, catalysts, pressure, initial temperature, and flow field. This book presents the fundamental bases of energetics of materials, deflagration and detonation, thermochemical process of decomposition and combustion, and combustion wave structures. The combustion mechanisms of various types of energetic materials, propellants, and explosives are discussed based on the heat transfer process in the combustion waves. The burning rate models are also presented to understand the rate-controlling steps of combustion processes, which show the relationships of burning rate versus pressure and initial temperature.

The combustion processes of propellants and explosives are highly complicated because of the heterogeneous structures of the materials, and because of phase changes from solid to liquid, and to gas under high pressure and temperature conditions. Therefore the real combustion phenomena are demonstrated by the use of a number of colored photographs during burning. New applications of energetic materials such as pyrolants and ducted rockets for an advanced propulsion system are also described.
